The absolute easiest way to get more power is to buy a gt, or cobra. If your car doesn't have rims/nice paint/cool looking hood/ 8.8 rear with gears... then I highly suggest buying a gt or cobra. I know a mustang can have lots of sentimental value, but ummm buying a gt would be the best route to take.... if money is an extreme object, other than that I'm with you all the way in building a bad v6.
A good daily driver set up would be this: CAI, 56mm throttle body, 5-speed, 8.8 with 3.73 or 4.10 gears, underdrive pulleys, long tuber headers, aluminum driveshaft, and lower control arms. That set up would at least get you in the low 15's. You would be suprised what a single port can do.... you would also be suprised in what it cannot do... bwhahaha.
Also, make sure your car has a rear spoiler, the Sn95 just doesn't look right without one. An Sn95 without a spoiler looks just like a 1979 smokey and the bandit trans am firebird... with no spoiler... which also adds to looking really fugly.
Remember, your V6 can hit 12's with less horsepower than a V8, but it takes alot more money to get there reliably.
A great set up would be this for a single port N/A + future supercharged or turbo'd V6:
4.3 stroker, with forged pistons+ crank, polished and ported heads, Fully ported/polished upper and lower single port intake that is cut and welded, custom cam, underdrive pulleys, 56mm throttle body, long tube headers, CAI.
This engine setup + gears, aluminum driveshaft, and lower control arms should get you into the low 13's. Possibly even into high 12's with drag radials, but I seriously doubt high 12's without forced induction.
With a supercharger, and gears you can easily expect mid 12's. YES even with the single port. Don't even worry about going split port. With the built 4.3+forced induction, you should be able to make it to the 11's, possibly even the 10's with lots of weight reduction and the right suspension set up/ drag radials.
Now, with a 347 stroker with the right H/C/I set up, and suspension, you can easily hit 10's N/A. It will also be cheaper to go the route of the 347 V8. Remember, horsepower doesn't mean crap if you can't get your power to the ground.
Going the V6 route will not be cheap unless you can find a Supercoupe engine, which I highly suggest since you can easily hit 12's with that motor. I've seen those engines sell for $300 dollahs.

Oh yeah tk1, I forgot to mention, the set up you mentioned would be bad ass, especially with gears, you would most definetly be in the 12's.
